SILVIA NEWELL DIRECTOR, MICHIGAN SEA GRANT (MISG) PROFESSOR, SCHOOL OF THE ENVIRONMENT AND SUSTAINABILITY UNIVERSITY OF MICHIGAN 400 NORTH INGALLS ST, ANN ARBOR, MI 48109 PHONE: 734-647-8215 SENEWELL@UMICH.EDU 3 PROJECT BACKGROUND THE NATIONAL SEA GRANT COLLEGE PROGRAM WAS ESTABLISHED BY U.S. CONGRESS IN 1966 (AMENDED IN 2008, PUBLIC LAW 110-394) TO BE A HIGHLY LEVERAGED FEDERAL AND STATE PARTNERSHIP TO HARNESS THE INTELLECTUAL CAPACITY OF THE NATION'S UNIVERSITIES AND RESEARCH INSTITUTIONS TO SOLVE PROBLEMS AND GENERATE OPPORTUNITIES IN COASTAL COMMUNITIES. FOR OVER 50 YEARS, THE NATIONAL SEA GRANT COLLEGE PROGRAM HAS SUPPORTED COASTAL AND GREAT LAKES COMMUNITIES THROUGH RESEARCH, EXTENSION, AND EDUCATION. SEA GRANT'S MISSION IS TO ENHANCE THE PRACTICAL USE AND CONSERVATION OF COASTAL, MARINE AND GREAT LAKES RESOURCES TO CREATE A SUSTAINABLE ECONOMY AND ENVIRONMENT. PROJECT DETAILS EACH UNIVERSITY-BASED SEA GRANT PROGRAM CONTRIBUTES TO THE NATIONAL FOCUS AREAS AND GOALS, AND MICHIGAN SEA GRANT IS NO DIFFERENT; WE ALIGN OUR FOCUS AREAS WITH THE NATIONAL SEA GRANT STRATEGIC PLAN'S FOCUS AREAS OF: HEALTHY COASTAL ECOSYSTEMS, SUSTAINABLE FISHERIES AND AQUACULTURE, RESILIENT COMMUNITIES AND ECONOMIES, AND ENVIRONMENTAL LITERACY AND WORKFORCE DEVELOPMENT. ACTIVITIES WITHIN EACH OF THESE FOCUS AREAS INCLUDE RESEARCH, EXTENSION AND EDUCATIONAL PROJECTS THAT AIM TO FURTHER THE NATIONAL SEA GRANT COLLEGE PROGRAM'S STATED MISSION. ANTICIPATED OUTCOMES AND RESULTS NOAA PROVIDES FUNDING TO SEA GRANT INSTITUTIONS TO INCREASE THE UNDERSTANDING, ASSESSMENT, DEVELOPMENT, MANAGEMENT, UTILIZATION, AND CONSERVATION OF THE NATION'S OCEAN, COASTAL, AND GREAT LAKES RESOURCES. THESE IMPORTANT EFFORTS PROMOTE A STRONG EDUCATIONAL BASE, RESPONSIVE RESEARCH AND TRAINING ACTIVITIES, BROAD AND PROMPT DISSEMINATION OF KNOWLEDGE AND TECHNIQUES, AND MULTIDISCIPLINARY APPROACHES TO ENVIRONMENTAL PROBLEMS, IN ACCORDANCE WITH 33 U.S.C. 1121(B). SPECIFIC OUTCOMES IDENTIFIED BY THE RECIPIENT INCLUDE, BUT ARE NOT LIMITED TO: STAKEHOLDERS HAVE ACCESS TO DATA, MODELS, POLICY INFORMATION, AND TRAINING THAT SUPPORT ECOSYSTEM-BASED PLANNING, DECISION MAKING, AND MANAGEMENT APPROACHES. THE SEAFOOD INDUSTRY INCLUDING CHARTER, COMMERCIAL, AND AQUACULTURE BUSINESSES IS KNOWLEDGEABLE ABOUT INNOVATIVE TECHNOLOGIES, APPROACHES, AND POLICIES. COMMUNITIES HAVE DIVERSE, HEALTHY ECONOMIES AND INDUSTRIES THAT ENHANCE NATURAL RESOURCE ASSETS WITHOUT DISPLACING TRADITIONAL WORKING WATERFRONTS. EDUCATORS ARE KNOWLEDGEABLE OF THE BEST AVAILABLE SCIENCE ON THE EFFECTIVENESS OF FORMAL AND INFORMAL EDUCATION ABOUT ENVIRONMENTAL SCIENCE. | Department of Health and Human Services | IND-ENABLING STUDIES OF AN ANTISENSE OLIGONUCLEOTIDE THERAPY FOR CLN3 BATTEN DISEASE - ABSTRACT GENOMIC DELETION OF EXONS 7 AND 8 FROM THE CLN3 GENE IS THE MOST COMMON CAUSE OF CLN3 BATTEN DISEASE (OMIM #204200), A RARE AUTOSOMAL RECESSIVE, LETHAL, NEURODEGENERATIVE LYSOSOMAL STORAGE DISORDER. THE FUNCTION OF THE PROTEIN IS NOT KNOWN, BUT THE LOSS OF ITS FUNCTION AND, POTENTIALLY THE CONSEQUENT PRODUCTION OF A TOXIC PROTEIN FRAGMENT, IS ASSOCIATED WITH ACCUMULATION OF AUTO-FLUORESCENT STORAGE MATERIAL AND GLYCEROPHOSPHODIESTERS (GPDS) IN CELLS, AND PROGRESSIVE VISION LOSS, COGNITIVE DECLINE, MOTOR COORDINATION DYSFUNCTION, SEIZURES, DYSTONIA AND ULTIMATELY DEATH IN CHILDREN AND YOUNG ADULTS. THE MEDIAN LIFESPAN OF THOSE WITH CLN3 DISEASE IS 20-30 YEARS. DR. HASTINGS AND THE TEAM INVOLVED IN THIS PROPOSAL HAVE DEVELOPED AN ANTISENSE OLIGONUCLEOTIDE (ASO) MEDICINE FOR TWO CHILDREN WITH CLN3 DISEASE CAUSED BY A DIFFERENT VARIANT, C.569DUPG, WHICH CAUSES AN OPEN READING FRAME SHIFT RESULTING IN A PREMATURE TERMINATION CODON (PTC). THE ASO INDUCES EXON SKIPPING OF EXON 8 TO REMOVE THE FRAMESHIFT AND RESCUE CLN3 PROTEIN EXPRESSION, ALBEIT VIA AN ALTERNATIVE, NATURALLY OCCURRING ISOFORM. THE CHILDREN WERE TREATED WITH THIS ASO BY INTRATHECAL INJECTION AND ARE RESPONDING REMARKABLY WELL TO THIS ASO THERAPY, CALLED ZEBRONKYSEN, EXHIBITING A REDUCTION IN DISEASE SYMPTOMS AND GPDS IN THE CEREBROSPINAL FLUID. ON THE SUCCESS OF THIS FIRST FRAME-SHIFT-CORRECTING ASO MEDICINE, WE PROPOSE NOW TO DEVELOP A SIMILAR APPROACH FOR THE MORE COMMON CLN3 DISEASE-CAUSING VARIANT CLN3EX7/8, WHICH HAS A PREMATURE TERMINATION CODON IN EXON 9. IN THIS CASE, WE HAVE DEVELOPED AN ASO THAT INDUCES SKIPPING OF EXON 5, WHICH RESULTS IN A NATURALLY OCCURRING IN-FRAME, CLN3 ISOFORM, CLN3EX5/7/8. WE HAVE GENERATED EXTENSIVE PRE-CLINICAL, PROOF-OF-CONCEPT EVIDENCE THAT CONVERSION OF CLN3EX7/8 TO CLN3EX5/7/8 EITHER GENETICALLY, IN A KNOCK-IN MOUSE MODEL OR USING ASOS IN PIG AND MOUSE WITH THE CLN3EX7/8 MUTATION, RESULTS IN LOWER DISEASE BURDEN. THIS PROJECT AIMS TO ESTABLISH THE SAFETY AND PHARMACODYNAMIC PROFILE OF THE ASO DRUG, SUPPORT ITS MANUFACTURING, AND PREPARE AN INVESTIGATIONAL NEW DRUG APPLICATION IN PREPARATION FOR A CLINICAL TRIAL TO INITIALLY TREAT TWO CHILDREN WITH THE DISEASE, WITH A LONG-TERM GOAL OF CREATING AN ASO THERAPY FOR ALL CHILDREN WITH THE MOST COMMON CAUSE OF CLN3 BATTEN DISEASE. | $1,347,673 |
