IF THE NEEDED SCIENCE DOESN'T EXIST, MNSG SEEKS TO FUND IT AND WORKS VIA EXTENSION, EDUCATION, AND COMMUNICATIONS TO ENSURE THAT SCIENCE GETS TO MINNESOTA COMMUNITIES. MNSGS OMNIBUS APPLICATION SEEKS TO SECURE FUNDING THAT WILL ENABLE MNSG STAFF AND SUPPORTED RESEARCHERS TO PRODUCE APPLICABLE SCIENCE, OUTREACH, EDUCATION, AND COMMUNICATIONS PRODUCTS AND TOOLS THAT INCREASE THE UNDERSTANDING, ASSESSMENT, DEVELOPMENT, MANAGEMENT, USE, AND CONSERVATION OF MINNESOTAS LAKE SUPERIOR AND INLAND COASTAL RESOURCES. ACTIVITIES TO BE PERFORMED: MNSG PROPOSES TO DEVELOP AND IMPLEMENT PRODUCTS AND TOOLS THAT INCLUSIVELY ADDRESS THE NEEDS OF THE COMMUNITIES WE SERVE. ACTIVITIES INCLUDE: (1) HOSTING A BIENNIAL REQUEST-FOR-PROPOSALS (RFP) COMPETITION AND FAST-TRACK RFP FOR RESEARCH THAT WILL ADDRESS THE NEEDS OF THE COMMUNITIES MNSG SERVE. (2) PRODUCING WEBSITES, WEB APPS, MULTIMEDIA, REPORTS, DISPLAYS/EXHIBITS, MEDIA RELATIONS, AND SCIENCE COMMUNICATION TRAINING THAT COMMUNICATE THE WORK OF MNSG STAFF AND SUPPORTED RESEARCHERS. (3) PRODUCING FISH-IDENTIFICATION MATERIALS FOR LAKE SUPERIOR CHARTER CAPTAINS AND FISH ECOLOGY EDUCATIONAL MATERIALS FOR GREAT LAKES EDUCATORS. (4) PRODUCING INFORMATION TO SUPPORT AND CREATE NETWORKS OF PEOPLE ENGAGED WITH LOCAL EMERGENCY SPILL RESPONSE NETWORKS. (5) PRODUCING MNSGS WATERSHED GAME THAT TEACHES ABOUT PRACTICES, PLANS, AND POLICIES THAT IMPROVE THE QUALITY OF A STREAM, LAKE, RIVER OR COASTAL REGION AND INCREASE RESILIENCE TO COMMUNITY FLOODING. (6) PRODUCING INFORMATION AND EDUCATIONAL MATERIALS ABOUT MAINTAINING SHORELINE ECOSYSTEMS AND SEPTIC SYSTEMS FOR HOMEOWNERS. (7) PRODUCING INFORMATION, TOOLKITS, AND HOW-TO GUIDES ON HOW COMMUNITIES CAN IMPLEMENT RESILIENCE PRACTICES. (8) PRODUCING K-12 LESSON PLANS ABOUT GREAT LAKES SHIPPING TO GREAT LAKES EDUCATORS. (9) HOSTING EVENTS AND PRODUCING INFORMATION TO INCREASE AQUACULTURE LITERACY IN MINNESOTA COMMUNITIES. MNSG WILL USE COASTAL RESILIENCE FUNDING TO SUPPORT RESILIENCE EXTENSION AND RESEARCH PROJECTS. EXPECTED OUTCOMES: (1) MNSG-SUPPORTED RESEARCHERS WILL PRODUCE OUTCOMES THAT ADDRESS THE NEEDS AND WILL BE USED BY THE COMMUNITIES MNSG SERVES. (2) MNSG COMMUNICATION PRODUCTS AND SERVICES WILL BE ACCESSED AND USED. (3) MNSG FISH IDENTIFICATION MATERIALS WILL BE USED BY CHARTER BOAT CAPTAINS AND THEIR CLIENTELE AND CONTRIBUTE TOWARD REDUCING ILLEGAL FISH HARVESTING. (4) MNSG FISH ECOLOGY EDUCATIONAL MATERIALS WILL BE USED TO IMPROVE GREAT LAKES FISHERIES LITERACY. (5) COORDINATION OF A GREAT LAKES-WIDE NETWORK OF PEOPLE KNOWLEDGEABLE ABOUT COMMUNITY SPILL RESPONSE WILL INCREASE THE NUMBER OF COMMUNITY MEMBERS WHO CAN ANSWER THE QUESTION: WHAT HAPPENS IF THERES A SPILL IN MY COMMUNITY? (6) MNSG WILL INCREASE THE NUMBER OF PEOPLE WHO USE INFORMATION FROM THE WATERSHED GAME TO MAKE SCIENCE-BASED WATER-MANAGEMENT DECISIONS. (7) MNSG MATERIALS ABOUT SHORELINE AND SEPTIC BEST PRACTICES WILL BE USED BY THE COMMUNITIES WE SERVE. (8) MNSG RESILIENCE INFORMATION (TOOLKITS, HOW-TO GUIDES) WILL BE USED IN THE COMMUNITIES WE SERVE TO PREPARE FOR AND RESPOND TO FLOODING AND OTHER EXTREME WEATHER EVENTS. (9) EDUCATORS WILL USE THE MNSG GREAT LAKES SHIPPING 101 LESSON PLANS. (10) MINNESOTA COMMUNITIES, INCLUDING LEGISLATORS, ARE MORE AWARE OF AQUACULTURE AND CONSUMERS CAN MAKE INFORMED DECISIONS ABOUT SEAFOOD PURCHASES. | Department of Health and Human Services | 700 MHZ NMR CONSOLE AND PROBE UPGRADE FOR BIOMOLECULAR AND 19 F NMR STUDIES AT THEUNIVERSITY OF MINNESOTA - THIS SHARED INSTRUMENTATION GRANT APPLICATION REQUESTS FUNDS TO UPGRADE THE 700 MHZ NMR SPECTROMETER AT THE MINNESOTA NMR CENTER (MNMR), A UNIVERSITY OF MINNESOTA CORE FACILITY THAT SERVES AS A HUB FOR HIGH-FIELD NMR SPECTROSCOPY. THE EXISTING INSTRUMENT IS CURRENTLY EQUIPPED WITH A 22-YEAR-OLD BRUKER AVANCE I CONSOLE AND TXI CRYOPROBE THAT ARE NOW OBSOLETE, INCREASINGLY UNRELIABLE, AND NO LONGER CAPABLE OF SUPPORTING MANY STATE-OF-THE- ART EXPERIMENTAL EXPERIMENTS. HARDWARE FAILURES, INCLUDING CRYOPLATFORM INSTABILITY, AUTO-TUNING ISSUES, AND CONSOLE COMMUNICATION PROBLEMS, HAVE SIGNIFICANTLY REDUCED INSTRUMENT AVAILABILITY AND LIMITED ITS UTILITY, EVEN AS USER DEMAND CONTINUES TO GROW. THE MANUFACTURER NO LONGER GUARANTEES PART AVAILABILITY OR TECHNICAL SUPPORT FOR THIS AGING PLATFORM, POSING AN ONGOING RISK TO RESEARCH CONTINUITY. THE PROPOSED UPGRADE INCLUDES A BRUKER AVANCE NEO CONSOLE AND QCI-F CRYOPROBE. THIS COMBINATION WILL RESTORE THE INSTRUMENT TO FULL OPERATIONAL CAPACITY AND PROVIDE A SIGNIFICANT TECHNICAL LEAP IN SENSITIVITY, STABILITY, AND EXPERIMENTAL FLEXIBILITY. SPECIFICALLY, THE UPGRADED SYSTEM WILL DELIVER IMPROVEMENTS IN 13C SENSITIVITY, ENABLE SIMULTANEOUS 1H DECOUPLING DURING 19F DETECTION, AND SUPPORT 19F-DETECTED EXPERIMENTS. THESE CAPABILITIES WILL OPEN NEW EXPERIMENTAL AVENUES FOR INVESTIGATORS STUDYING CONFORMATIONAL EXCHANGE, LIGAND SCREENING, PROTEIN- SMALL MOLECULE INTERACTIONS, INTRINSICALLY DISORDERED PROTEINS, AND MACROMOLECULAR DYNAMICS ON THE ΜS–MS TIMESCALE. IN ADDITION, THE SYSTEM WILL SUPPORT AUTOMATION FEATURES, AS WELL AS COMPATIBILITY WITH TOPSPIN 4.X SOFTWARE AND THE EXISTING AUTOSAMPLER, FURTHER IMPROVING EFFICIENCY, REPRODUCIBILITY, AND THROUGHPUT. MNMR CURRENTLY SUPPORTS 32-36 ACTIVE RESEARCH GROUPS EACH YEAR, WHOSE WORK RESULTS IN 30-40 PEER-REVIEWED PUBLICATIONS ANNUALLY AND IS SUPPORTED BY 16-20 NIH-FUNDED PROJECTS. THE USER BASE SPANS FIVE COLLEGES AND INCLUDES RESEARCHERS FROM MORE THAN A DOZEN DEPARTMENTS ACROSS THE TWIN CITIES CAMPUS. CURRENT USERS APPLY NMR TO A BROAD SPECTRUM OF BIOMEDICAL PROBLEMS, INCLUDING DNA REPAIR, CANCER SIGNALING, INFECTIOUS DISEASE, METABOLISM, AND DRUG DISCOVERY. THE MNMR CENTER IS ALSO AN IMPORTANT TRAINING RESOURCE, PROVIDING HANDS-ON EXPERIENCE IN HIGH-FIELD NMR METHODS FOR GRADUATE STUDENTS, POSTDOCTORAL FELLOWS, AND TECHNICAL STAFF. THE REQUESTED UPGRADE WILL ALLEVIATE THE SIGNIFICANT PRESSURE CURRENTLY PLACED ON THE CENTER’S TWO 600 MHZ INSTRUMENTS, WHICH ARE NEARING FULL CAPACITY DUE TO USERS SHIFTING AWAY FROM THE OUTDATED 700 MHZ INSTRUMENT. BY RESTORING THIS UNDERUTILIZED INSTRUMENT TO FULL FUNCTIONALITY AND ENABLING ADVANCED BIOMOLECULAR AND SMALL MOLECULE NMR EXPERIMENTS, THE UPGRADE WILL ENHANCE ACCESS, EXPAND CAPACITY, AND IMPROVE DATA QUALITY FOR A BROAD AND GROWING COMMUNITY OF NIH-FUNDED INVESTIGATORS. THIS INVESTMENT WILL NOT ONLY STRENGTHEN ONGOING RESEARCH PROGRAMS BUT ALSO ATTRACT NEW USERS TO THE MNMR CENTER. THUS, THIS PROPOSAL ADDRESSES AN URGENT INSTRUMENTATION NEED AND WILL HAVE IMMEDIATE AND LASTING IMPACT ON NIH-SUPPORTED RESEARCH ACROSS STRUCTURAL BIOLOGY, CHEMICAL BIOLOGY, AND BIOMEDICAL SCIENCES AT THE UNIVERSITY OF MINNESOTA. | $1,334,250 |
